การศึกษาความแม่นยำในการวินิจฉัยแบบหลายศูนย์ (multicenter diagnostic accuracy study) ประเมินว่าการทดสอบดัชนี (index test) เช่น การตรวจหาชีวมาร์เกอร์ การถ่ายภาพทางการแพทย์ หรือกฎการทำนายทางคลินิก สามารถระบุภาวะเป้าหมาย (target condition) ได้ดีเพียงใดเมื่อดำเนินการในศูนย์ปฏิบัติการทางคลินิกตั้งแต่สองแห่งขึ้นไปที่เป็นอิสระต่อกัน การรวบรวมผู้ป่วยจากสภาพแวดล้อมที่หลากหลายทำให้ได้ค่าประมาณความไว (sensitivity) ความจำเพาะ (specificity) และอัตราส่วนความน่าจะเป็น (likelihood ratios) ที่มีความเที่ยงตรงภายนอก (externally valid) มากกว่าที่ได้จากการศึกษาศูนย์เดียว และช่วยให้สามารถประเมินได้อย่างชัดเจนว่าประสิทธิภาพของการทดสอบแตกต่างกันอย่างไรในแต่ละศูนย์ กลุ่มผู้ป่วย และระดับทักษะของผู้ปฏิบัติงาน
