Semen Quality Evaluation

Semen Quality Evaluation and Breeding Soundness Assessment · Also known as: breeding soundness examination, seminal analysis, sperm quality assessment

Semen quality evaluation is a systematic assessment of male animal reproductive capacity, measuring sperm characteristics and overall breeding soundness. Developed by veterinary andrologists in the 1970s, the practice combines objective measures—sperm concentration, motility, morphology—with functional tests to predict fertility potential. Evaluation is essential for identifying suitable breeding animals, managing reproductive health, and ensuring successful artificial insemination (AI) programs.

When to use it

Semen evaluation is essential when establishing AI programs, certifying animals for breeding soundness, investigating subfertility, monitoring reproductive health, and screening breeding candidates. Use before purchasing breeding animals, if conception rates decline, or when managing high-value genetics. Assumptions include proper sample collection and storage, trained microscopists, and standardized protocols. Prefer objective measures and CASA analysis for consistency.

Species-dependent: Bull semen typically ≥50 million sperm/mL with ≥60% progressive motility; boar semen ≥50 million/mL with ≥70% motility; ram semen ≥100 million/mL with ≥70% motility. Standards vary by breed and individual baseline.

What percentage of sperm abnormalities is acceptable?

Most breeding standards allow ≤30% total morphological defects (head, midpiece, tail abnormalities combined). Some defect types (e.g., proximal cytoplasmic droplets) are more tolerable than others (e.g., acrosome defects).

How does semen quality change with age?

Sperm production peaks in young adults and gradually declines with age. Concentration and motility may remain relatively stable, but morphological defects increase. Older males may require more frequent evaluation.

Why is temperature control critical in semen handling?

Sperm are temperature-sensitive. Cold shock (rapid cooling below 15°C) or heat stress damages the cell membrane and reduces motility. Samples must be maintained at or above ambient temperature until analysis.
