Seed Germination Test

Standard Seed Germination Testing and Viability Assessment · Also known as: Germination assay, Seed vigor test, Seed quality evaluation

Seed Germination Test is an analytical and physiological pipeline for assessing seed viability and germination rate under controlled conditions. Standardized by ISTA (International Seed Testing Association), this method quantifies the proportion of seeds capable of normal seedling development and informs seed quality certification, planting decisions, and storage management.

When to use it

Use this test before purchasing or planting seed. Mandatory for seed companies and certified seed producers. Essential if seed has been stored >1 year, exposed to unfavorable conditions (heat, humidity), or if field establishment is critical (high-value crops). Optional for home-saved seed if previous years showed consistent good germination.

  • Distinguishes dead from dormant seeds, informing pre-treatment needs (cold stratification, priming).

Limitations
  • Laboratory conditions (constant temperature, moisture) may not reflect field conditions; some seeds passing germination tests fail in cold, wet soil.
  • Does not assess seed vigor traits (vigor = capacity to perform well under stress); two lots with 90% germination may differ in field establishment.
  • Dormancy can inflate apparent germination failure; warm-season seeds may show low germination if tested immediately after harvest.
  • Disease-infected seeds may pass germination tests but carry pathogens that affect later growth.

What germination percentage is acceptable for planting?

Standards vary by crop: cereals typically ≥85–90%, oilseeds ≥90%, vegetables ≥75–95%. Acceptable levels also depend on economic risk and field conditions; if establishment is risky (wet, cold soil), target >90% to ensure adequate stand. Always adjust seeding rate: desired stand = target seedlings / (germination × seed vigor factor).

How do I know if low germination is due to dormancy rather than seed death?

Apply tetrazolium (TZ) staining to non-germinated seeds: living tissue (with active metabolism) stains pink/red; dead tissue remains colorless. If TZ-positive seeds are >10% of the non-germinated fraction, dormancy is likely; pre-treatment (warm water soak, cold stratification) may improve germination. For dead seeds, the lot should be rejected or used with increased seeding rate.

Can I do a simple germination test at home without a laboratory?

Yes, using the paper towel or sand method: roll moist paper towels with seeds, keep at room temperature 20–25°C, and count emerged radicles at 5–7 days. This gives approximate germination but may not exactly match ISTA results (protocol differences). Use for quick screening; send official samples to an ISTA lab for certification or disputes.

How long can I store seeds while maintaining germination?

Seed longevity depends on species, moisture content, and temperature. Orthodox seeds (wheat, maize) store well: at 10–15°C and 5–8% moisture, can last 5–10 years. Recalcitrant seeds (cocoa, avocado) lose viability in months. Store in cool, dry, dark conditions. Test germination every 1–2 years if relying on stored seed.
