Differential analyse af enkeltcelle-RNA-sekventering

Differential analyse af enkeltcelle-RNA-sekventering (scRNA-seq) er en beregningsmæssig pipeline, der sammenligner transkriptomiske profiler på tværs af biologiske betingelser – såsom behandlet versus ubehandlet, sygdom versus sundhed eller tidspunkter – med opløsning på enkeltcelle-niveau. Den identificerer, hvilke gener, celletyper og cellestater der ændrer sig mellem betingelserne, og giver mekanistisk indsigt, som bulk-RNA-seq-sammenligninger ikke kan tilbyde. Tilgangen kombinerer klyngedannelse, celletypeannotation og statistisk testning, typisk ved brug af pseudobulk-aggregering for at tage højde for korrelation inden for prøver.
