单细胞序列比对 — scRNA-seq 读段比对

单细胞序列比对是计算步骤,它将单细胞 RNA 测序实验产生的数百万个短测序读段映射回参考基因组或转录组。与批量 RNA 测序比对不同,每个读段都带有一个细胞条形码和一个唯一分子标识符 (UMI),它们共同识别起始细胞和单个 RNA 分子。准确的比对和条形码解复用是构建驱动所有下游单细胞分析的细胞-基因计数矩阵的先决条件。
