Developed systematically by Mordenti and colleagues in the late 1980s, it enables rational first-in-human dose prediction without assuming species-specific metabolic differences.PBPK is a mechanistic modeling framework that uses physiological parameters, tissue properties, and drug-specific attributes to predict drug concentration time profiles in the body. Developed rigorously in the 1990s by researchers including Nestorov, PBPK integrates anatomy, biochemistry, and kinetics to enable rational drug development, bridging in vitro data to clinical outcomes.
