ResuméAllometric scaling is a mathematical approach for predicting human pharmacokinetics from preclinical animal data using body weight relationships. Developed systematically by Mordenti and colleagues in the late 1980s, it enables rational first-in-human dose prediction without assuming species-specific metabolic differences.Michaelis-Menten kinetics describes the rate of enzyme-catalyzed reactions as a function of substrate concentration. Developed by Leonor Michaelis and Maud Menten in 1913, this foundational framework models enzyme catalysis through the rapid-equilibrium approximation and enables prediction of drug metabolism rates in pharmacokinetics.
