Introduced by Amidon and colleagues in the 1995 Biopharmaceutics Classification System, it bridges laboratory measurements and clinical outcomes to streamline drug development.PBPK is a mechanistic modeling framework that uses physiological parameters, tissue properties, and drug-specific attributes to predict drug concentration time profiles in the body. Developed rigorously in the 1990s by researchers including Nestorov, PBPK integrates anatomy, biochemistry, and kinetics to enable rational drug development, bridging in vitro data to clinical outcomes.
